Glucocorticoid Receptor Polymorphism A3669G Is Associated with Airflow Obstruction in Mild-to-Severe Asthma

چکیده

Background: Glucocorticoids (GCs) represent the mainstay therapy for asthmatics. A subset of severe asthmatics fails to respond steroid-based therapies, leading important healthcare costs. Single nucleotide polymorphisms (SNPs) glucocorticoid receptor genes were associated with a response GC. We evaluate possible relation BclI and A3669G SNPs clinical, biological functional characteristics Methods: recruited 172 mild-to-severe asthmatic outpatients referring Severe Asthma Rare Lung Disease Unit at San Luigi University Hospital. Clinical data obtained recruitment when spirometry tests peripheral blood sampling performed. Patients genotyped through pyrosequencing assay results. Results: AG genotype younger, allergic had higher IgE levels compared AA (p < 0.05). Moreover, lower post-bronchodilator FEV1/FVC ratio than GG 0.05), RV/TLC Conclusions: The might be related type-2 asthma; in particular, allele SNP was GC our In conclusion, this observational cross-sectional study suggests role as predictor asthma severity phenotype.

Parameters associated with persistent airflow obstruction in chronic severe asthma.

The significance of severe airflow obstruction in severe asthma is unclear. The current study determined whether severe airflow obstruction is related to inflammatory or structural changes in the airways. Patients with severe asthma from a tertiary referral clinic were divided into two groups according to their postbronchodilator forced expiratory volume in one second (FEV1): severe persistent ...

Common Genetic Variant of insig2 Gene rs7566605 Polymorphism Is Associated with Severe Obesity in North India

Background: Obesity is a very common disorder resulting from an imbalance between food intake and energy expenditure, and it has a substantial impact on the development of chronic diseases.  The aim of this study was to examine the association of INSIG2 (rs7566605) gene polymorphism with obesity and obesity associated phenotypes in North Indian subjects. Methods: The variants were investig...

Factors associated with persistent airflow limitation in severe asthma.

Persistent airflow limitation can develop in nonsmoking patients with asthma. However, the prevalence and risk factors for airways obstruction with incomplete reversibility in asthma are unknown. We assessed the prevalence of persistent airflow limitation (defined as postbronchodilator FEV(1) or FEV(1)/VC < 75% predicted) in 132 nonsmoking outpatients with severe asthma visiting chest physician...

Glucocorticoid receptor-binding characteristics in severe asthma.

The cellular mechanisms associated with severe asthma are still poorly understood. This study investigated the association between glucocorticoid-receptor (GR) alterations and continuous oral glucocorticoid therapy requirement in severe asthma. GR-binding affinity (Kd) and receptor number (n) in peripheral blood monocytes (PBM) obtained from 10 normal subjects, 10 untreated, intermittent asthma...

ETB receptor polymorphism is associated with airway obstruction

BACKGROUND Endothelin-1 (EDN1) has been involved in the development of airway obstruction and inflammation in asthma. Several polymorphisms have been identified among the genes encoding for preproET1, an inactive precursor of ET-1, and for ETA (EDNRA) and ETB (EDNRB), the two receptors for EDN1. In the present work, we hypothesised that molecular variation in these genes could be a major determ...
